Opto2P-FCM: a MEMS-based miniature two-photon microscope with two-photon patterned optogenetic stimulation Journal Article uri icon

Overview

abstract

  • ; Miniature two-photon (2P) microscopes capable of imaging and; cell-specific photostimulation are important tools for all-optical; interrogation of neural circuits in behaving animals. The Opto2P-FCM; achieves this through a dual-path design that combines high-resolution; two-photon laser-scanning microscopy at 920 nm using a; micro-electro-mechanical systems (MEMS) mirror with a separate; 1030 nm beam path that delivers spatially patterned; photostimulation through a coherent fiber bundle, which also collects; fluorescence.; In vivo; , the system; supports stable functional imaging during locomotion and optogenetic; activation of targeted cells. The work demonstrates a compact; architecture for precise optical stimulation and imaging in a single; head-mounted device, with applications in neuroscience for detailed; studies of how specific patterns of neural activity drive; behavior.;
